ABSTRACT

Objective:To optimize the alcohol extraction process of herbal pair puerarin-berberine.Methods:Based on the etha-nol reflux extraction, the extraction quantity of total flavonoids, total alkaloid, puerarin and berberine hydrochloride were used as the e-valuation index, and the independent variables included the drug particle, ratio of solid to liquid, ethanol concentration, reflux dura-tion and reflux times. Significance analysis was evaluated by Plackett-Burman design, and then the extraction process was optimized by Box-Behnken response surface methodology. Results:The optimal extraction conditions of drug pair puerarin-berberine were as follows:the drug particle was 80 mesh, the ratio of solid to liquid was 1:13,the ethanol concentration was 75%,the reflux time was 60 min, and the reflux times was 4. Under the above conditions, the extraction quantity of total flavonoids, alkaloid, puerarin and berberine hydrochloride was 120.34,56.99,109.63 and 39.26 mg ·ml-1, respectively. Conclusion: The extraction process of herbal pair puerarin-berberine is reasonable and feasible optimized by Plackett-Burman design and Box-Behnken response surface methodology.

ABSTRACT

Objective:To evaluate the effect of pinus massoniana bark extract (PMBE) and grape seed extract (GSE) on dentin demineralization caused by acid.Methods:40 root dentin blocks with half of the surface covered were randondy divided into 4 groups (n =10).All samples were subjected to pH cycling for 8 days, and deionized distilled water(DDW),0.1%NaF,12% PMBE solution and 12% GSE solution were used as the experimental solutions in the 4 groups. The dentin mineral density(DMD) of the both sides was determined using micro-computed tomography. The D-value of DMD between nn-demineralized and demineralized side (△DMD)was calculated. The samples were observed with field emission scanning electron microscops (FE-SEM).Results:The △DMD of DDW,NaF,PMBE and GSE groups was 198.64 ±59.97,45.94 ±24.21,90.23 ±28.77 and 105.07 ±29.53 respectively. The △DMD between PMBE and GSE groups had no significant difference (P > 0.05),which were both higher than that of NaF group (P < 0.05) and lower than that of DDW group(P < 0.05).The FE-SEM revealed that the dentin tubules in DDW group were completely open, but in NaF group were essentially closed in PMBE group and GSE group were spindle shaped or narrow crack opening. Conclusion:PMBE and GSE had almost the same effect on improving the acid resistance of dentin.

ABSTRACT

Objective · To investigate effect of fucoidan on autophagy, migration and invasion in human multiple myeloma U266 cells. Methods · The U266 cells treated with fucoidan were cultured in vitro. The formation of autophagosomes was observed by transmission electron microscopy (TEM). Transwell assay was used to evaluate the effect of fucoidan on migratory and invasive abilities of U266 cells. The protein levels of LC3-Ⅱ/LC3-Ⅰ, Beclin-1, P62, MMP9, CXCR4, p-AKT/T-AKT, p-mTOR/T-mTOR were detected by Western blotting. MMP9 concentration in the culture medium was examined by ELISA. Results · ① Autophagosomes increased in fucoidan-treated cells compared with control group under TEM. ② Migratory and invasive abilities were inhibited by fucoidan in a dose-dependent manner, which were suppressed by chloroquine. ③ Western blotting demonstrated that expression of LC3-Ⅱ/LC3-Ⅰ, Beclin-1 and MMP9 increased in fucoidan-treated cells, while P62, CXCR4, p-AKT/T-AKT and p-mTOR/T-mTOR decreased compared with control group. ④ The result of ELISA showed that MMP9 concentration in the culture medium of fucoidan-treated cells significantly decreased. Conclusion · Fucoidan induces autophagy and inhibits migration and invasion in U266 cells.

ABSTRACT

Objective:To prepare ketoprofen-loaded solid lipid nanoparticles ( SLN) and evaluate the properties. Methods: The formula was optimized by orthogonal experiments with the encapsulation efficiency as the index. The optimal preparation was investiga-ted by the morphology, particle size, zeta potential and drug forms. The release property was characterized by a dialysis method and the release process was fitted. Results: The best formula was as follows: poloxamer 0. 1g, Tween-800. 2g, lecithin 0. 15g, glycerol monostearate 0. 05g and ketoprofen 50mg. The particles were spherical in shape with the encapsulation efficiency of 61. 95%, the par-ticle size was 151. 7 nm and the zeta potential was-30. 2 mV. The result of DSC indicated the drug dispersed in the lipid matrix was a-morphous and molecular state. The in-vitro release curve showed the release was rapid at the early stage and then slowed down with the accumulated amount up to (85. 11 ± 7. 62)% in 12h. The drug was released slowly from SLN with the matrix erosion. The release pro-file fitted well with a Higuchi equation. Conclusion: The solid lipid nanoparticles containing ketoprofen exhibit good quality and the preparation method is simple and feasible, therefore, it is valuable to be further studied.

ABSTRACT

Objective:To analyze and optimize the extraction technology of Geum aleppicum by Box-Behnken design and response surface methodology. Methods:With the content of total flavonoids as the index, the effects of three factors including the solvent-solid ratio, ethanol concentration and extraction time on the content of total flavonoids were studied by single factor experiment, and the ex-traction technology was optimized by Box-Behnken design and response surface methodology. Results: The optimal extraction condi-tions of Geum aleppicum were as follows:the solvent-solid ratio was 4. 2, the ethanol concentration was 50% and the extraction time was 80 min. Under the above conditions, the average yield was 12. 590 0 mg·g-1 . Conclusion: Optimizing extraction technology of the total flavonoids in Geum aleppicum by Box-Behnken design and response surface methodology is reasonable, and the mathematical mod-el is consistent with the experimental data, which has good predictability.

ABSTRACT

Objective To investigate the differences of regional homogeneity(ReHo) in depressed patients with vary degrees of anxiety using resting-state function MRI,and the relationship vary degrees of anxiety symptoms. Methods 10 depressed patients with scores less than 1 in anxiety/somatization factor of Hamilton depression scale,10 depressed patients with scores above 2,and 10 age-,gender-,education-matched healthy controls were underwent resting-state fMRI scanning at 3.0 Tesla.Correlation analysis between ReHo with significant differences level of brain region and the total score of Hamilton depression scale, and anxiety/somatization factor were underwent respectively. Results The brain regions with significant difference level among three groups were the left posterior cerebellum(-39,-77,-29 ; K =15),the left angular gyrus (-56,-63,22 ; K =13) and the left anterior cingulate (-3,22,29 ; K =19) (P < 0.05,Alphasim) ; compared with healthy group, the mild anxiety group showed significantly decreased ReHo in the left posterior cerebellum, and increased ReHo in the left anterior cingulate; the moderate and severe anxiety group showed significantly decreased ReHo in the left posterior cerebellum, and increased ReHo in the left anterior cingulate and the left angular gyrus; no significant differences were found in patient groups. In the mild anxiety group, the left anterior cingulate positively correlated with the anxiety severity(r=0.646,P=0.043),and in the severe anxiety group positively correlated with the somatic anxiety(r =0.693,P=0.038).Conclusion Depressed patients showed abnormal neural activity in resting state, and the degree of abnormality is associated with clinical anxiety/somatization symptoms.
